Culture Media

  • For reviving freeze-dried cultures, the following liquid media without agar are used.
    PDA ‘BD Difco’, Corn meal extract powder ‘Nissui’ (CMA), Standard Method Agar ‘Nissui’ (YPDA), King B Agar ‘Eiken’ medium commercially sold is not applicable for preparing liquid culture, because it contains agar in its formula for solidification.
  • Media should be sterilized at 121°C for 15-20 minutes.
  • Media without pH indication are those for which pH adjustment is unnecessary.

media for filamentous fungi and yeasts

PDA (Potato Dextrose Agar)

PDA ‘BD Difco’ 39.0 g/l
Agar 3.0 g/l
applicable for most of filamentous fungi
Although PDA ‘BD Difco’ contains agar, 3.0g/l more agar could preferably be added to get a harder agar surface for better practice of microbial inoculation/transplantation

CMA (Corn Meal Agar)

Corn meal extract powder ‘Nissui’ 17.0 g/l
Agar 3.0 g/l
Although Corn meal extract powder ‘Nissui’ contains agar, 3.0g/l more agar could preferably be added to get a harder agar surface for better practice of microbial inoculation/transplantation
To prepare culture medium for marine fungi, 38.0g/l of dried seawater powder (A JAPAN PET DRUGS CO.,LTD.) could be added to it.

V-8 medium

  1. Mix 400 mL of commercially available V8 juice with 600 mL of distilled water and 4.5 g of CaCO3. After stirring, centrifuge the mixture.
  2. Add 800 mL of distilled water and 18.0 g of agar to 200 mL of the obtained supernatant.

SNA (Synthetic low-Nutrient Agar)

LCA (Miura medium)

KCl 0.2 g/l
MgSO4・7H2O 0.2 g/l

IFO-7 medium

EBIOS powder 5.0 g/l

IFO-9 medium

YM (Yeast Malt peptone agar)

YPGS(Yeast extract Peptone Glucose Saccharose)

medium for growth and investigation Liquid medium for bacteria other than root nodulating bacteria, primarily used for reviving freeze-dried cultures.

YPDA(Yeast-extract Peptone Dextrose Agar)

Standard Method Agar ‘Nissui’ 23.5 g/l
Agar 3.0 g/l
medium for most bacteria.
Though Standard Method Agar ‘Nissui’ contains agar, we add 3.0g/l more agar to it in order to stiffen it and inoculate easier.

medium for Bradyrhizobium

Yeast extract 0.4 g/l
Mannitol 10.0 g/l
K2HPO4 0.5 g/l
MgSO4・7H2O 0.2 g/l
NaCl 0.1 g/l
Agar 18.0 g/l
pH 6.8
for root nodulating bacteria, Bradyrhizobium.For Rhizobium, Add 3.0 g of CaCO3 to this medium.

TTC(2,3,5-Triphenyl Tetrazolium Chloride)

Peptone 10.0 g/l
Casein hydrolysate 1.0 g/l
Glucose 5.0 g/l
1% solution of TTC 1ml g/l
Agar 15.0 g/l
medium for Ralstonia solanacearum.TTC solution (filtered through a 0.2µ filter) should be added at 50-60℃ after autoclaving a mixture of other components.

MRS (de Man-Rogosa-Sharpe)

MRS medium ‘OXOID’ 52.0 g/l
Agar 18.0 g/l
for lactic acid bacteria.

King B

King B Agar ‘Eiken’ 38.0 g/l
Glycerin 10.0 g/l
King B medium is a confirmation medium for the identification of fluorescein-producing Pseudomonas.

YMA (Yeast Malt extract Agar)

Malt extract 10.0 g/l
Glucose 4.0 g/l
Agar 18.0 g/l
pH 7.3
for most Actinomycetes.

Albumin medium

Albumin medium 0.25 g/l
Glucose 1.0 g/l
K2HPO4 0.5 g/l
MgSO4・7H2O 0.2 g/l
1% solution of Fe2(SO4)3 1 ml
Agar 18.0 g/l
pH 6.8-7.0
medium for growth of genus Streptomyces.
