Instruction for Transfer of Slant Cultures

Manipulate following procedures as carefully and aseptically as possible, using a clean bench. Moreover, store the slants in the refrigerator when strains are not transferred immediately.

  1. Prepare an instrument consisting of Nichrome wire, the tip of which is bent like a hook, and fix it to a handle with a metal stick and a plastic grip (hereinafter referred to as the “platinum wire”) as shown in figure.

  2. Pass the openings of the test tubes of the distributed slants and the new ones to which strains are transferred, through the flame of a gas burner or alcohol lamp. (Do not to heat the silicon plugs too much, or they may burn.)

  3. Burn well the tip of the platinum wire shown in 1 and the part to which the Nichrome wire is attached. In particular, heat the Nichrome wire until the wire is red hot to sterilize it.

  4. Open the lids of the new plate agar media or the silicon plugs of the new slant media. Let the tip of the heated platinum wire touch the agar until the wire is cooled down sufficiently.

  5. Remove the silicon plug of the distributed slant culture strains. Scratch the agar, including the mycelia block, with the tip of the platinum wire cooled as shown in 4. Transfer the block onto a fresh plate or slant media. While the agar block including the mycelia is transferred, it must not be touched the inside surfaces of the culture tubes, especially the rim.

  6. Reheat over the flame the rims and the tips of the silicon plugs of donor and recipient culture tubes. Replace the plugs while they are hot. (Be careful not to touch the heated part.) Close the cover rapidly in the case of transferring to plate media.
